梅花鹿茸蛋白质酶法水解研究

摘要 目的研究梅花鹿茸蛋白质酶法水解条件。方法用胰酶、胰蛋白酶、木瓜蛋白酶在相同条件下水解梅花鹿茸蛋白质,比较氨基氮生成率,选择适宜酶类。用所选定的酶在不同温度(T)、不同底物浓度(S)、不同酶与底物浓度比(E/S)和不同水解时间(t),水解梅花鹿茸蛋白质,以氨基氮生成率为指标确定最佳水解条件。结果胰蛋白酶为适宜酶,最佳水解条件为:温度45℃、pH8.0、底物浓度7.5%、酶与底物浓度比6000u/g、时间6h。结论胰蛋白酶是水解梅花鹿茸蛋白质较适宜的酶,在上述最佳条件下,水解梅花鹿茸蛋白质,氨基氮生成率达43.52%,氮溶解指数达93%,水解液中蛋白质绝大多数为低肽分子,更有利于机体消化吸收。 Objective To study protein enzymatic hydrolysis conditions from sika pilose antler. Methods Sika pilose antler protein was hydrolyzed with trypsin, pancreafin and papain under the same condition, amino nitrogen production rate was compared, suitable enzyme was choose. The protein enzymatic hydrolysis in sika pilose antler was researched with different substrate concentration, enzyme/substrate ratio, and hydrolysis time. The optimum parameters of reaction were determined with amino nitrbgen as the index. Resuits Trypsin is suitable for protein hydrolysis in sika pilose antler. The results showed that the optimun conditions of hydrolysis are the temperature at 45℃, the original pH 8.0, substrate concentration 7.5%, enzyme/substrate ratio ( E/S ) 6000 u/g, and hydrolysis time 6h. Conclusion Trysin is suitable enzyme for protein hydrolysis in sika pilose antler. Under optimun conditions, amino nitrogen production rate is up to 43.52%, the index of soluble nitrogen is up to 93% in hydrolysate, The most of protein in hydrolysate is low molecular weight peptide, which is good for digestion and absorption of human body.
