摘要
目的探讨屋尘螨1类变应原(DerP1)对嗜酸性粒细胞(E)释放炎性细胞因子IL-1β、IL-6、IL-8、IL-10和TNF-α的影响。方法用CD16抗体磁珠法从新鲜人外周血分离E;5×105/mlE与5μg/mlDerP1共培养12h;用流式细胞小球微阵列(cytometricbeadarray,CBA)法定量细胞培养上清液中细胞因子。结果外周血E单独培养过程中仅有极少量IL-1β、IL-6、IL-8、IL-10和TNF-α释放,但与5μg/mlDerP1共育时,培养上清液中上述5种细胞因子浓度显著增加(P均<0.001),IL-10增加约5倍,其他4种细胞因子均增加约100倍。结论DerP1对E释放炎性细胞因子具有很强的促进作用。
Objective To investigate the role of Dermatophagoides pteromyssims 1 (Der p1 ), the allergen of house dust mite, in release of inflammatory cytokines including interleukin (IL)-1β, IL-6, IL-8, IL-10 and tumor necrosis factor (TNF)-α from activated eosinophils. Methods Eosinophils were purified from fresh human bully coat by magnetic cell sorting with anti-CD16 magnetic beads. The purified eosinophils(5×10^5/ml) were incubated with Der pl (5 μg/ml) for 12 h. The cytokines in the supernatant of cultured eosinophil were determined by microarray technique ( Cytometric Bead Array, CBA). Results The releases of IL-1β, IL-6, IL-8, IL-10 and TNF-α were significantly increased in the supernatant of eosinophils cultured with Der pl (5 μg/ml) compared with those of eosinophils alone ( all P 〈 0. 001 ). Conclusion The allergen of house dust mite Der pl can activate purified human eosinophils to induce release of inflammatory cytokines.
