摘要
目的在原代培养的新生大鼠心肌细胞上,观察丹参酮ⅡA(TSN)对血管紧张素Ⅱ(AngⅡ)诱导的心肌细胞肥大的影响。方法应用细胞培养技术培养新生大鼠心肌细胞,采用相差显微镜测量细胞大小,测定心肌细胞3H-亮氨酸掺入作为心肌细胞肥大的指标;用流式细胞仪测定心肌细胞凋亡率,用Western blot法检测心肌细胞凋亡蛋白P53、Bax和Bcl-2的表达。结果TSN能抑制AngⅡ诱导的心肌细胞增大、心肌细胞3H-亮氨酸掺入率的显著增加(P<0.01)、心肌细胞凋亡率的增加(P<0.05)、心肌细胞促凋亡蛋白P53和Bax表达的明显增高(P<0.01),其效果与AngⅡ受体阻滞剂缬沙坦(Valsartan)相似。结论TSN可以抑制AngⅡ诱导的心肌细胞肥大,这可能与其同时抑制了心肌细胞的凋亡有关。
[Objective] To investigate the effect of Tanshinone Ⅱ A (TSN) on the hypertrophy induced by Angiotensin Ⅱ(Ang Ⅱ ) in the primary culture of neonatal rat cardiomyocytes. [Methods] Following the index of cardiomyocyte hypertrophy, the cell size was determined by phase contrast microscope and ^3H-Leucine incorporation was investigated; The rate of cardiomyocyte apoptosis was analysed with flow cytometric analysis. The P53, Bax and Bcl-2 expression was assessed using Western blot. [Results] After Ang Ⅱ treatment, the cell size and 3H-Leucine incorporation rate of cardiomyocyte and P53, Bax expression of cardiomyocytes increased significantly. TSN inhibited these effects of Ang Ⅱ, and the effects of TSN was similar to Valsartan, which is the inhibitor of angiotensin Ⅱ receptor. [Conclusion] TSN can prevent the hypertrophy of cardiomyocytes induced by Ang Ⅱ, which may relate to the depress of cardiomyocyte apoptosis by TSN.
