摘要
目的探讨CpGODN体外活化的外周血单个核细胞(PBMC)后对肿瘤细胞的杀伤效应。方法CpG-ODN体外刺激人外周血单个核细胞(PBMC),将活化的PBMC与肿瘤细胞按50∶1的比例共孵育72h后,以MTT和酶学检测活化的PBMC对肿瘤细胞的杀伤作用。结果CpG-ODN诱导活化的PBMC对肿瘤细胞的杀伤作用增强,而对正常肝细胞则无明显的杀伤作用。结论CpG-ODN可诱发机体免疫效应,提高对肿瘤细胞的杀伤作用。
[Objective] To study the antitumor effect of human peripheral blood mononuclear cells(PBMC) activated by synthetic oligodeoxynucleotides(ODN) containing unmethylated CpG dinucleotides(CpG-ODN) in vitro. [Methods] CpG-ODN was used to stimulate PBMC. PBMC activated by CpG-ODN was co-cultured with QSG7701, HepG2, K562, A549 and 5-8F cells respectively for 72 h. MTT method and enzyme assays in the supernatant were uesd to detect PBMC-mediated antitumor activity. [Results] CpG-ODN enhanced antitumor activity of PBMCs, but it didn't up-regulate the PBMC killing activity to QSG7701 cells. [Onclusion] The study indicates that, as a novel immunoregulator, CpG-ODN can enhance antitumor activity by stimulating PBMC in vitro.
