摘要
目的:探讨烧伤血清作用下中性粒细胞CD11/CD18和内皮细胞ICAM-1表达的变化。方法:流式细胞术检测烧伤病人伤后第1、3、5、7天多核中性粒细胞(PMN)CD11a/CD18和CD11b/CD18的表达和烧伤血清刺激后人脐静脉内皮细胞(HUVEC)ICAM-1的表达,并应用免疫胶体金银染色观察烧伤血清刺激PMN后1、3、6、12、24h表达CD11a/CD18和CD11b/CD18情况。结果:烧伤病人伤后第一天PMN膜表面CD11a/CD18和CD11b/CD18表达明显升高达峰值,并在伤后一周内维持此高水平状态。烧伤血清刺激正常人PMN后1h。CD11a/CD18和CD11b/CD18表达迅速增高达峰值,24h内仍维持此高水平。而烧伤血清作用于培养良好的HUVEC后,ICAM-1表达逐渐增多,12h达高峰。结论:烧伤早期PMN激活而表达CD11a/CD18和CD11b/CD18,烧伤血清可分别刺激PMN和HUVEC表达CD11a/CD18、CD11b/CD18和ICAM-1,从而导致烧伤后PNN与内皮细胞粘附,对PMN在各脏器中滞留和组织损伤起作用。
Objective: To observe the changes of expression of CD11/CD18 on polymorphonuclear neutrophils (PMNs) and ICAM-1 on endothelial cells (ECs) under the stimulation of burn serum. Methods: Cytometry was employed to detect the expression of CD11a/CD18 and CD11b/CD18 on PMNs on the 1st, 3rd, 5th and 7th postburn day in burn patients and that of ICAM-1 on human umbilical vein endothelial cells (HUVECs) after the stimulation with burn serum. Meanwhile, immunogold-silver staining was used to observe the expression of CD11a/CD18 and CD11b/CD on PMNs on the 1st, 3rd, 6th, 12th and 24th hour after the stimulation with burn serum. Results: ① The expression of CD11a/CD18 and CD11b/CD18 on PMNs was rapidly increased to the peak value on the 1st postburn hour in burn patients and remained the same within one week; ② The expression of CD11a/CD18 and CD11b/CD18 on PMNs from healthy individuals was also rapidly increased to the peak value on the 1st hour after the stimulation with burn serum but remained the same for only 24 hours; ③ The expression of ICAM-1 on well-cultured HUVECs was gradually increased and reached its peak value on the 12th hour after the stimulation with burn serum. Conclusion: In the early stage after burns, PMNs were activated to express CD11a/CD18 and CD11b/CD18. Burn serum can respectively stimulate PMNs to express CD11a/CD18 and CD11b/Cd18 and HUVECs to express ICAM-1, which might result in PMN-EC adhesion.
