兔眼房水及玻璃体中川芎嗪含量的高效液相色谱法测定 被引量:3

摘要 目的用高效液相色谱法测定兔眼房水、玻璃体中川芎嗪的含量,了解川芎嗪注射液直流电离子导入后的有效成分在兔眼房水和玻璃体中的吸收、分布、代谢及排泄情况。方法色谱柱为Waters公司symmetryC18(5μm,4.6×150 mm),流动相:甲醇∶水=40∶60,1000 ml中含SDS0.9130 g,磷酸二氢钾0.6770 g,冰醋酸1 ml;检测波长:294 nm,流速:1 ml/min,室温操作。结果在1.25~36μg/ml范围内,房水中川芎嗪平均回收率均在80%左右,日内精密度在5%左右,日间精密度在10%左右;在0.3125~10μg/ml范围内,玻璃体中川芎嗪平均回收率均在80%左右,日内精密度及日间精密度均在10%左右。结论当川芎嗪浓度在一定范围内时,兔眼房水和玻璃体中川芎嗪峰面积与浓度呈良好线性关系。 OBJECTIVE To determine the content of ligustrazine in rabbit aqueous humor and vitreous with high performance liquid chromatography and to understand the absorption,distribution,metabolism and excretion of direct current of transmitting the ion of active component.METHODS The analytical column was a symmetry C18 stainless steel column(5μm,4.6×150mm),water methanol(60∶40),in 1000ml mixture of the mobile phase there are 0.9130g SDS,0.6770g KH2PO4 and 1ml Glacial acetic acid.The flow rate is 1.0ml/min.The wavelength of the detector is 294nm.RESULTS Wen concentration of liqustrazine is in the range of 1.25~36μg/ml in aqueous humor.The average recovery was 80%,RSD of intra-day was 5%,while inter-day was 10%.When concentration of liqustrazine is in the range of 0.3125~10μg/ml in vitreous.The average recovery was 80%,RSD of intra-day and inter-day was 10%.CONCLUSIONS The relationship between the peak area and concentration of the drug is linear well with the concentration in some invariably range.
