摘要
目的:探讨缺氧诱导因子-1α(HIF-1α)、环氧化酶-2(COX-2)及CD105在人脑胶质瘤组织中的表达及与临床病理特征的关系。方法:应用免疫组化SP法检测70例胶质瘤和10位正常人脑组织中HIF-1α、COX-2和CD105的表达,并测定CD105标记的MVD。结果:1)胶质瘤组织中HIF-1α、COX-2和CD105蛋白的阳性表达率分别为64.3%、61.4%和74.3%,明显高于正常人脑组织。2)HIF-1α、COX-2表达和MVD与胶质瘤病理分级密切相关。3)胶质瘤组织中HIF-1α和COX-2蛋白表达的一致符合率为70.35%,且两者表达呈正相关,r=0.5837,P=0.004。4)HIF-1α和COX-2阳性表达组的MVD(35.87±4.55和36.00±4.57)均高于阴性表达组(29.19±4.69和29.48±4.71),P值均为0.0000;两者均为阳性表达时的MVD(36.22±4.62)最大,明显高于其他各组(33.75±3.59和28.29±4.33),P=0.0001;两者表达均与MVD呈正相关,r值分别为0.5405和0.4652,P值分别为0.005和0.014。结论:HIF-1α、COX-2和CD105过表达可能在胶质瘤的恶性进展中发挥重要作用,并与肿瘤血管形成密切相关,三者的联合检测可更准确地揭示胶质瘤的生物学特性。
OBJECTIVE: To investigate the relationship be tween the expressions of HIF-1α, COX-2 and CD105 in human gliomas and clinicopathological features. METHODS:The expressions of HIF-1α, COX-2 and CD105 were examined by SP immunohisto chemical staining in 70 human glioma specimens and 10 normal human brain tissue specimens, and MVD labeled by CD105 were measured. RESULTS: 1) The positive expression rates of HIF-1α, COX-2 and CD105 proteins in gliomas (64.3%,61.4%,74.3%) were significantly higher than those of normal human brain tissues. 2) The expressions of HIF-1α, COX-2 and MVD were associated with the pathological grading of gliomas. 3)The expression conformation rate of HIF-1α and COX-2 was 70.35%, and there was a positive correlation between the expressions of HIF-1α and COX-2, r=0.583 7,P=0.004. 4)The MVD values in HIF-1α and COX-2 positive groups [(35.87 ± 4.55) ; (36.00±4.57)] were significantly higher than those in negative groups, respectively [(29. 19±4.69) ;(29.48±4.71)],and P was both 0.000 0, The MVDvalue (36.22±4.62) in both HIF-1α positive and COX 2 positive group was significantly higher than those in the other subgroups [(33.75±3.59),(28.29±4.33)],P=0.000 1, and the expressions of them were all positively correlated with CD105- MVD, r=0.540 5,P=0.005;r=0.465 2,P=0.014. CONCLUSION:The overexpressions of HIF-1α, COX-2 and CD105 may play an important role during the malignant progression of glioma and they were closely associated with tumor angiogenesis, so their cojoined de tection may better reveal biological characteristics of glioma.
