摘要
本实验通过PCR-SSCP技术检测了三大品种6个类型共296匹马ELA-DRA* exon2的多态性。用12%非变性聚丙烯酰胺凝胶电泳将已变性的PCR扩增产物分离,并用银染法显色。结果296匹马中共出现6种基因型:三种纯合子分别记为AA、BB和CC型,均为3条带;三种杂合子分别记为AB、BC和AC型,均为4条带;且AA和AB两种基因型是所研究的各类型马的优势基因型。纯血马和三河马的PIC和He值均较高,属于高度多态,4个类型蒙古马均属于中度多态。聚类分析表明三大品种各聚一支,各类型蒙古马中锡尼河马和巴尔虎马关系较近。
ELA - DRA * exon2 locus was used to detect 296 horses from 6 types of three big breeds. The PCR amplified products of the locus were detected by 12 percent non - denatured polyacry lamide gel electrophoresis and showing in silver staining protocol. The results showed that 6 genotypes were detected in 296 horses: 3 homozygotes named AA, BB and CC, respectively and they all had 3 bands; 3 heterozygotes named AB, BC and AC, respectively, and they all had 4 bands; AA and BB were dominant genotypes in all horses. The values of He and PIC in Thoroughbred horse and Sanhe horse represent highest polymorphism, 4 horse types of Mongolian belonged to intermediate polymorphism. Clustering analysis showed that three big breeds gathered one branch respectively, Xinihe horse and Baerhu horse had nearly relation among Mongolian horse.
