摘要
目的:探讨RASSF1A、CyclinD1和Bcl-2蛋白在膀胱移行细胞癌发生发展中的作用,以及RASSF1A和CyclinD1与Bcl-2蛋白之间的关系。方法:免疫组化法(SP法)检测45例膀胱移行细胞癌组织、45例癌旁黏膜及18例正常黏膜组织中RASSF1A、CyclinD1和Bcl-2蛋白的表达。结果:膀胱移行细胞癌组织中RASSF1A、CyclinD1和Bcl-2蛋白表达阳性率分别为46.7%(21/45)、88.9%(40/45)和57.8%(26/45);癌旁组织分别为84.4%(38/45)、42.2%(19/45)和24.4%(11/45);正常组织分别为94.4%(17/18)、38.9%(7/18)和16.7%(3/18)。膀胱移行细胞癌组织中RASSF1A蛋白的表达显著低于癌旁及正常膀胱黏膜,P<0.05;而CyclinD1和Bcl-2蛋白的表达则显著升高,P<0.05。RASSF1A和CyclinD1及Bcl-2蛋白的表达呈负相关性,P<0.05。结论:RASSF1A蛋白的表达降低和CyclinD1、Bcl-2蛋白的过表达可能参与膀胱移行细胞癌的发生发展过程,RASSF1A可能通过某种机制促使CyclinD1和Bcl-2在膀胱移行细胞癌中过表达,从而促进膀胱移行细胞癌的发生发展。
OBJECTIVE:To study the expressions of RASSF1A, Cyclin D1 and Bcl-2 in transitional cell carcinoma of bladder, and study the relationship among RASSF1A protein and Cyclin D1 protein, Bcl-2 protein. METHODS:The expressions of RASSF1A protein, Cyclin D1 protein and Bcl 2 protein in transitional cell carcinoma of blad der, mucous in the paracarcinoma and normal mucous of urinary bladder, were detected by the immunohistochemical staining SP method. RESULTS: The positive expressions of RASSF1A protein,Cyclin D1 protein and Bcl-2 protein by immunohistochemistry were 46.7%(21/45), 88.9% (40/45) and 57.8% (26/45) in transitional cell carcinoma of bladder, 84.4% (38/45), 42.2% (19/45), 24.4% (11/45) in paracarcinoma mucous, 94.4%(17/18),38.9%(7/18), 16.7% (3/18) in normal mucous of urinary bladder, respectively. The expression of RASSF1A protein was significant lower in transitional cell carcinoma of bladder tissues than those in paracarcinoma mucous and normal mucous of urinary bladder, P〈0. 05. The expression of Cyclin D1 protein and Bcl-2 protein were significantly higher in transitional cell carcinoma of bladder tissues than those in paracarcinoma mucous and normal mucous of urinary bladder, P〈0. 05. There was a negative correlation among RASSF1A protein and Bcl-2 protein, Cyc- lin D1 protein in transitional cell carcinoma of bladder tissues, P〈0.05. CONCLUSIONS: The abnormal expressions of RASSF1A protein, Cyclin D1 and Bcl-2 protein maybe have highly correlation with the oneo- genesis and development of bladder cancer. RASSF1A protein may increase the expression of Cyclin D1 protein and Bcl-2 protein in some ways, and then lead to the oncogenesis and development of bladder cancer.
