摘要
目的:探讨建立在超声导引下大鼠经皮肝内乙醇注射(PEI)的模型,并动态观察肝功能恢复特点。方法:取SD大鼠40只,在超声引导下确定肝中叶后,注射无水乙醇0.1 m l,分别在第1、3、5、7 d取血,检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)含量,并取注射部位肝组织做病理组织学检查。结果:PEI术后第1 d大鼠血清ALT、AST的含量升高最明显,随后开始下降,第7 d恢复术前水平。大体观察及HE病理染色均可显示肝组织部分坏死。结论:在超声导引下对大鼠肝脏进行乙醇注射,其乙醇局限性扩散可导致肝细胞坏死,是研究PEI治疗肝脏疾病的可靠动物模型。
Objective:To develop a mouse model of injecting ethanol into the liver under ultrasonographic guidance and study its liver function dynamically. Methods : Forty SD rats were performed percutaneous ethnol injection into the liver under ultrasonographic guidance. The AST and ALT were determinded prior to the surgery and 1,3,5 and 7 d after surgery. At the same time the morphological changes in the liver also observed. Resuhs:AST and ALT were increased most at the first day after PEI then decreased ,until the 7th day, it became to normal. HE stained and observed by eye,we can see part of the liver necrosis. Conclusions : Injecting ethanol into the mouse's liver under ultrasonographic guidance could induce high concentrated ethanol locally which cause the liver cells necrosed, This model might be valuable of researching the PEI treating liver disease.
