葡萄胎和绒毛膜癌组织中葡萄糖调节蛋白94的表达及意义 被引量:5

摘要 目的:探讨葡萄糖调节蛋白94(glucose-regulated protein,GRP94)在葡萄胎和绒毛膜癌组织中的表达及意义。方法:采用DA-KO EnVision免疫组织化学染色法对39例葡萄胎及20例绒毛膜癌组织中的GRP94进行染色,镜下观察GRP94在葡萄胎和绒毛膜癌组织中的特点,用图像分析软件对其积分光密度(IOD)进行分析。结果:39例葡萄胎组织中34例(87.2%)滋养层细胞胞质有GRP94表达,且合体滋养层细胞胞质比细胞滋养层细胞胞质表达强,P=0.000。在20例绒毛膜癌组织中16例(80%)癌细胞胞质有GRP94表达,同时也发现GRP94在合体滋养层细胞胞质比细胞滋养层细胞胞质表达强,P=0.017,且癌细胞胞质比癌旁平滑肌细胞胞质的表达强,P=0.000。在葡萄胎和绒毛膜癌滋养层细胞胞质中GRP94的表达随分化程度降低而升高,P=0.04;其中,在细胞滋养层细胞胞质中GRP94的表达也是随分化程度降低而升高,P=0.005。而在合体滋养层细胞胞质中发现GRP94的表达与分化程度不相关,P>0.05。结论:GRP94的表达强度与滋养细胞肿瘤的分化程度有关。 OBJECTIVE: To study the expression of glucose-regulated protein 94 (GRP94) in human hydatidiform mole and choriocarcinoma. METHODS: GRP94 expression was detected by DAKO EnVision immunohistochemistry, and its characters were analyzed by the microscope analysis system and integrated optical density (IOD) analysis software. RESULTS: Thirty-four of 39 cases (87.2%) of hydatidiform mole showed GRP94 positive staining, and GRP94 expression of syncytiotrophoblast cytoplasm was higher than that of cytotropho blast cytoplasm,P=0.000. Sixteen of 20 cases (80%) of choriocarcinoma showed GRP94 positive staining, GRP94 expres- sion of syncytiotrophoblast cytoplasm also was higher than that of cytotrophoblast cytoplasm, P=0. 017, and GRP94 expres sion in choriocareinoma was higher than that in its adjacent tis sues,P = 0. 000. All in all, the levels of GRP94 expression were correlated with cell differentiation in trophoblastic tumor (P= 0.04), and the levels of GRP94 expression were correlated with cell differentiation in cytotrophoblast (P= 0. 005). However, the expression of GRP94 and hydatidi have not significant differences in ehoriocarcinoma and hydatidiform mole syncytiotrophoblast,P〉0.05. CONCLUSION:The expression of GRP94 is correlated with the malignant change of gestational trophoblastic tumor.
