摘要
目的:研究西红花酸对大鼠血小板聚集的影响。方法:采用Born比浊法测定血小板聚集性,采用胶原黏附法测定血小板黏附率,以血流变仪和微量血浆测试装置测定全血和血浆黏度,以试剂盒测定乳酸脱氢酶(LDH)活性。结果:体内外实验表明,西红花酸(100、50、25mg·kg-1)有效抑制由4,5-腺苷二磷酸二钠盐(ADP)和胶原诱导的大鼠血小板聚集(P<0.01),最大抑制率分别为36.6%(ADP)和33.3%(胶原);对由花生四烯酸(AA)诱导的血小板聚集无作用(P>0.05);对大鼠血小板黏附、全血和血浆黏度以及LDH水平无明显影响(P>0.05)。结论:西红花酸对由ADP和胶原诱导的大鼠血小板聚集具有显著的抑制作用,对由AA诱导的大鼠血小板聚集无明显抑制作用,对血小板黏附亦无明显抑制作用。
AIM:To study the effect of crocetin on platelet aggregation in rats in vivo and in vitro. METHODS: The platelet aggregation was measured by Born-nephelometer. The platelet adhesion was measured with collagen adhesion. The whole blood and plasma hemorheological parameters were determined by viscometer. The activities of plasma lactate dehydrogenase (LDH) were assayed with kits. RESULTS: Compared with the control group, crocetin (100 , 50, 25 mg·kg-1) could significantly inhibit platelet aggregation induced by ADP and collagen(P〈0.01), and the maximum inhibition ratio were 36.6% (ADP) and 33.3% (collagen)respectively. But crocetin did not inhibit platelet aggregation induced by AA (P〉0.05). It couldn′t affect the platelet adhesion,the whole blood and plasma hemorheological parameters and plasma LDH levels. CONCLUSION: Crocetin can inhibit platelet aggregation induced by ADP and collagen, but has no effect on platelet aggregation induced by AA.
