摘要 目的研究左旋甲状腺素钠(Thyroxine,T4)对大鼠垂体-卵巢轴内分泌功能及形态的影响。方法采用放射免疫分析法和电子显微镜技术,系统观察T4对大鼠血清性激素含量及靶腺组织形态的影响。结果T40.1μg.g-1.d-1,腹腔注射,连续用药21d,可显著降低大鼠血清黄体生成素(LH)的水平(P<0.01),而对大鼠卵泡刺激素(FSH)、雌二醇(E2)、孕酮(P)的含量无显著影响。T40.6μg.g-1.d-1可明显降低雌性大鼠LH,E2,P的水平(P<0.01),对大鼠FSH的含量无显著性影响(P>0.05)。电镜观察显示,高剂量0.6μg.g-1.d-1,腹腔注射,21d组SD大鼠腺垂体促性腺细胞、卵巢内分泌细胞超微结构发生退行性改变。结论T4大剂量长期应用对雌性大鼠垂体-卵巢内分泌功能有抑制作用,此抑制作用可能与靶腺内分泌细胞超微结构的退行性改变有关。 Objective To study the effects of Levothyroxine(T4) on endocrine functions and morphous in pituitary-ovary axis ( POA ) in rats. Methods The serum hormone concentration was measured with radioimmunoassay (RIA) and the morphology of gonad endocrine cells was observed with electron microscope. Results T4 ( 0.1 μg · g^-1 d^-1, ig,21d) decreased serum LH concentration in female rats significantly( P 〈0.01 ) ,but had no effect significantly on serum FSIq, E2 and P concentration ( P 〉 0.05 ). T4 (0.6μg · g^-1· d^-1, ig, 21 d ) decreased serum LH, E2 and P concentration in female rats significantly(P 〈 0.01 ) , but had no significantly effects on serum FSH concentrations in female rats ( P 〉 0.05 ). Electron microscope observation showed that the obvious retrograde changes in the pituitary gonadotroph cells and ovarian endocrine cells, especially in T4 high dosage group. Conclusion High dose T4 long-term administration has significantly inhibitory effects on the secretory functions of the pituitary and ovary, the inhibitory effects are related to the retrograde changes of the uhramicrostructure in the target gland endocrine cell.
