摘要
目的探讨肿瘤侵袭相关基因EMP3和PCDH-γ-A11的mRNA在原发人脑胶质瘤组织中的表达情况,分析其表达变化与胶质瘤恶性程度的关系。方法分别应用传统的RT-PCR和SYBR Green实时定量PCR检测方法检测EMP3和PCDH-γ-A11基因mRNA在30例原发胶质瘤(WHO分级:Ⅱ级10例,Ⅲ级10例,Ⅳ级10例)和10例正常脑组织中的表达情况。统计学分析两个基因在胶质瘤和正常脑组织间的表达差异,并进一步结合临床资料分析不同分级、不同性别和不同年龄组间的表达差异。结果在各级别胶质瘤中EMP3和PCDH-γ-A11的mRNA与正常脑组织相比均表达下调,并具有显著意义(P<0.01);EMP3在Ⅱ级和Ⅳ级及Ⅲ级和Ⅳ级胶质瘤间的表达也存在统计学差异(P<0.05);PCDH-γ-A11在Ⅱ、Ⅲ和Ⅳ级胶质瘤相互之间的表达无统计学差异(P>0.05)。结论各级别胶质瘤中EMP3和PCDH-γ-A11的mRNA表达均显著低于正常脑组织,并随胶质瘤恶性程度的增加表达量逐渐降低。EMP3基因的表达下调与胶质瘤的恶性程度密切相关。
Objective To detect the mRNA expressions of tumor suppressor genes epithelial membrane protein 3 (EMP3) and protocadherin-gamma subfamily A11 (PCDH-γ-A11) in primary human brain glioma tissues, and analyze the relationship between the changes in the mRNA expressions and the degree of malignancy of gliomas. Methods The mRNA expressions of EMP3 and PCDH-γ-A11 in primary glioma tissues of 30 cases (WHO grading: a respective 10 cases of Grade Ⅱ, Ⅲ and Ⅳ ) and 10 normal brain tissues were detected by the conventional reverse transcription-polymerase chain reaction (RT-PCR) and SYBR Green quantitative real-time PCR, respectively. Differences in mRNA expressions of the two genes between gliomas and normal brain tissues were statistically analyzed, and the relationship between the mRNA expressions and clinical data was further analyzed by different pathological grades, gender and age. Results Expressions of EMP3 and PCDH-γ-A11 mRNA were all significantly down-regulated in gliomas of different pathological grades, compared with that in the normal brain tissues; differences in mRNA expressions between the gliomas and normal brain tissues had statistical significance (P〈0.01). Expressions of EMP3 mRNA between gliomas of Grade Ⅱ and Ⅲ, and Ⅲ and Ⅳ had statistical differences (P〈0.05). Expressions ofPCDH-γ-A11 mRNA in gliomas of Grade Ⅱ, Ⅲ and Ⅳ had no statistical differences (P〉0.05). Conclusion The mRNA expressions of the tumor suppressor genes EMP3 and PCDH-γ-A11 in gliomas of different pathological grades are obviously lower than that in normal brain tissues, and the expression levels decrease gradually with the increase in the degree of malignancy of gliomas. The down-regulation of EMP3 mRNA expression is closely related with the degree of malignancy of gliomas.
