摘要
目的:观察高碘对大鼠甲状腺功能及病理生理改变的影响。方法:以高碘喂养雌性SD大鼠,检测游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)水平,用半定量RT-PCR、Western印迹及免疫组织化学方法检测大鼠甲状腺细胞Trail的表达。结果:高碘组较对照组甲状腺功能增高(P<0.05)。半定量RT-PCR、Western印迹及免疫组织化学方法结果显示Trail在正常大鼠甲状腺细胞中有表达,在高碘组表达增高(P<0.01)。结论:高碘使大鼠甲状腺细胞Trail表达增高,可能通过引起Trail表达的增强诱导甲状腺细胞过度凋亡,进而影响其功能及病理生理改变。
Objective :To investigate the effects of excess iodine on thyroid function and pathologic changes of normal rats. Methods: Female SD rats were feed with high-iodine diets. Immunofluorescence was used to detect the free triiodothyr - onine ( FT3 ) and free thyroxine (FY4) in serum. Semi - quantity RT - PCR, Western - blotting and Immunohistochemical staining were used to detect the expression of Trail in rats. Results: The concentration of FT3 and FY4 increased in the high - iodine group compared to the control group ( P 〈 0.05 ). The expression of Trail was observed in thyroid gland of normal rats. Its expression in the high - iodine group was increased compared to the control group (P〈0.01 ). Conclusion: The expression of Trail was increased under the induction of excess iodine in rats. The diet with high iodine may induce the apoptosis of thyroid cells by increasing the expression of Trail, and further lead to functional and pathologic changes of thyroid zrand.
