摘要
采用微波辅助萃取和AB-8型大孔树脂从葛根中提取和纯化总黄酮。分别考察了乙醇浓度、固液比、微波功率和辐射时间对提取率的影响;吸附柱类型、进液浓度、进液量、进液流速和pH值对吸附的影响和洗脱剂乙醇浓度、洗脱剂流速和洗脱剂体积量对脱附的影响,并分别确定了它们适宜的工艺条件。在适宜工艺条件下,微波辅萃总黄酮提取率为83.94%,纯化后产品总黄酮纯度达70%。建立了AB-8型大孔树脂吸附葛根总黄酮的Freundlich等温吸附方程,模型与实验值基本相符。
Microwave-assisted extraction and AB-8 type of microporous resin were used to separate and purify the drug component-total isoflavones from Pueraria. The effects of ethanol concentration, ratio of solid to liquid, microwave power and radiation time on the extraction rate, the effects of resin column type,feed concentration,feed volume,feed flow rate and pH value on adsorption and the effects of eluant concentration,eluant volume and eluant flow rate on desorption were investigated and their suitable technical conditions were determined, under which the extraction rate and the total isoflavones purity of the purified product were 83.94% and 70% respectively.The Freundlich isothermal adsorption equation of AB-8 type resin was established which was basically in accordance with the experimental, data.
