摘要
【目的】研究缺血预处理对大鼠脑组织微粒体钙摄取能力的影响。【方法】采用四血管阻断法大鼠全脑缺血模型,将24只大鼠随机分为3组:手术对照组(n=8)、预处理缺血组(n=8)和脑缺血组(n=8),两步离心法提取脑组织微粒体,紫外法监测其对Ca2+的摄取。【结果】手术对照组和预处理缺血组的Ca2+最大摄量均显著高于脑缺血组(P<0.01)。【结论】缺血预处理可提高脑微粒体的Ca2+摄取能力,可能通过减轻钙超载而达到脑保护的作用。
[Objective] To observe the effects of ischemic preconditioning on the calcium uptake of cerebral microsomes. [Methods] The whole cerebral ischemia was induced with four-blood vessel occlusion. Twenty-four Wistar rats were randomly divided into three groups: operation control group ( n = 8), cerebral ischemic preconditioning group ( n = 8) and cerebral ischemla group ( n = 8). Cerebral microsomes were prepared with two-step method, and the Ca^2+ uptake was observed by UV spectrophotometer. [Results] The Ca^2+ uptake of operation control group and cerebral ischemic preconditioning group were both more than that of cerebral ischemia group ( P 〈 0.01 ). [Conclusion] The results suggest that significant improvement of ischemia preconditioning to Ca^2+ uptake in cerebral microsomes may be associated with the decrease of calcium overload and cerebral prevention.
