摘要
目的:探讨清心开窍方对老年痴呆模型小鼠脑内自由基代谢及海马CA1区神经细胞形态学的影响。方法:将150只小鼠随机分为五组,即空白对照组、模型组、模型+中药大剂量组(14.82 g·kg-1)、模型+中药小剂量组(7.41 g·kg-1)、模型+脑复康对照组(0.42 g·kg-1)。除空白对照组外,其余各组均每日灌服AlCl3造摸(200 mg·kg-1),用药各组自造模第7 d起,每日上午给予AlCl3灌胃,下午给予相应的药物,每日1次,连续8周,9周后评定疗效。测小鼠学习记忆功能和脑内自由基代谢指标(SOD、MDA、LF)及对海马CA1区神经细胞形态学观察。结果:中药各组能明显减少海马神经细胞的破坏,提高SOD活性,降低MDA及LF含量(P<0.01),学习记忆能力明显改善。结论:清心开窍方能改善AD小鼠学习记忆的能力,对海马神经细胞具有保护作用,提高痴呆小鼠的抗氧化能力,降低氧自由基对机体的损伤,而对AD具有很好治疗作用。
Objective:To study the effects of the Qing Xin KaiqiaoFang(QXKQF) on contents of free radicals metabolism in brain and shape hippocampal nerve cells in Alzheimer disease (AD) mice. Methods: One handred fifty mice were divid into five groups:blank group,model group, two groups of treatment b), QXKQF (14.82 g·kg^-1,7.41 g·kg^-1), Piracetan comparison group (0.42 g·kg^-1 ). The model group was orally given AlCl5 (200 mg·kg^-1 ) every day. For QXKQF and Piracetan groups, AlCl5 treatment was given 6 days at the begining, followed by giving orally AlCl5 in the morning and drug in the afternoon for 8 weeks. Then, learning and memory ability, free radicals metabolism in brain(SOD, MDA, LF) and morphology of hippocampal nerve cells were investigated. Results:QXKQF can prevent hippocampal nerve cells from damage obviously. The SOD increased, the contents of MAD and LF were lowered compared with model group( P 〈 0.01 ). Learning and memory ability of QXKQF groups is improved, compared with model group( P 〈 0.05 -0.01 ). Conclusion: QXKQF can improve learning and memory ability of AD mice, protect hippocampal nerve cells, resist oxidation and reduce free radicals injure for AD mice.
