摘要
采用纤维素酶法,研究了纤维素酶分离提取富硒灵芝菌丝体多糖的条件,以提高菌丝体多糖的得率。利用正交试验设计,确定了优化的提取条件,即:浸提加水量50mL/g菌丝体,酶解时间80min,加酶量300U/mL,pH4.5,温度55℃。在优化的实验条件下,菌丝体多糖的得率33.6%。利用QSepharose Fast Flow离子交换层析分离纯化菌丝体多糖,多糖的回收率达82.8%。
Extraction conditions of mycelium polysaccharides from selenium-rich Ganoderma lucidum were studied by cellulase for increasing of yield. The factors influencing extraction of mycelium polysaccharides such as water,time,enzyme,pH and temperature were studied by orthogonal experiment design. The optimum conditions were obtained, water 50 mL per gram mycelia,time 80 min,enzyme 300 U/mL,pH 4.5 and temperature 55 ℃. The yield of mycelium polysaccharides was 33.6% under the optimum conditions. Mycelium polysaccharides were purified by Q Sepharose Fast Flow ion exehange chromatography and the recovery was up to 82.8%.
