姬松茸多糖通过增强小鼠脾淋巴细胞功能诱导Bel-7402细胞凋亡 被引量:1

摘要 目的:探讨姬松茸多糖(ABP)对Bel-7402细胞凋亡的影响及其机制。方法:制备ABP与小鼠脾淋巴细胞共培养上清(ABP-SL-CM),作用于Bel-7402细胞,流式细胞仪分析诱导凋亡情况以及p53基因的蛋白表达;透射电子显微镜观察细胞形态学变化。结果:不同浓度的ABP-SL-CM能诱导Bel-7402细胞凋亡,流式细胞仪检测凋亡率高达21.68%,DNA直方图上可见到亚二倍体峰,透射电子显微镜显示细胞凋亡的典型形态学变化;p53蛋白的荧光强度值组间比较无显著性差异(P>0.05)。结论:ABP通过增强小鼠脾淋巴细胞活性诱导Bel-7402细胞凋亡,但其诱导凋亡机制未见与p53基因蛋白表达有关。 Objective: To investigate the effect and the mechanism of Agaricus blazei Muill Polysaccharides (ABP) on Bel -7402 cell apoptosis. Methods: Flow cytomertric analysis were used to detemine the apoptotic cells and intracellular tumor inhibitive gene p53 expression; Electron microscope was applied to observe the morphological change. Results: when ABP - SL - CM treated Bel - 7402 cells for 48 hours in vitro, the ABP - SL - CM were found to induce cell apoptosis; Flow cytomertric analysis revealed that 21.68% ( ABP - SL - CM - L) apoptotic cells were seen while in control culture (N - SL - CM) there were only few(8. 14% ) apoptotic cells; The expression of intracellular p53 expression had not apparently changed( P 〉 0. 05 ). Conclusion: ABP can induce cell apoptosis, the mechanism was related to enhancing mice splenic lymphocytes activity and not be proved to be related to p53 expression.
