榄香烯对人肺癌细胞表面所表达与侵袭转移相关黏附蛋白的影响 被引量:7

摘要 目的:观察榄香烯对肺癌细胞系表面所表达与侵袭、转移相关的黏附蛋白的影响。方法:利用激光共聚焦显微镜观察高转移人巨细胞肺癌细胞PG和低转移人肺腺癌细胞PAa表达的血小板免疫相关抗原CD42a、TSP、CD9的表达情况;利用流式细胞仪检测的高转移人巨细胞肺癌细胞PG和低转移人肺腺癌细胞PAa表达的血小板免疫相关抗原CD42a、TSP、CD9的表达情况及二种癌细胞与高、中、低浓度的榄香烯注射液共同孵育后CD42a、TSP、CD9的表达改变。结果:利用激光扫描可见CD9、CD42a、TSP均匀的表达于细胞表面。流式细胞术结果显示,PG细胞表达的CD9阳性细胞数目和平均荧光强度低于PAa细胞,而PG细胞表达的CD42a、TSP阳性细胞数目和平均荧光强度均高于PAa细胞,从总的趋势来看,榄香烯注射液增高PG和PAa细胞CD9的表达,降低其CD42a、TSP的表达。结论:PG细胞系的转移能力高于PAa细胞系;对肺癌细胞表面所表达与侵袭、转移相关的粘附蛋白的影响,可能是榄香烯注射液抗肺癌转移的分子机理。 Objective :To observe the influence of Elemene in the invasion and metabasis related surface adhesive proteins on lung cancer cell. Methods : We used confocal microscopy observed the expression of the thrombocytic immune related antigen CD42a,TSP and CD9 on high metastasis lung cancer cell line PG and low metastasis lung cancer cell line PAa. We also used flow cytometer observed the expression of the thrombocytic inlnlune related antigen CD42 a, TSP and CD9 on PG and PAa,and the alter of the expression of CD42a,TSP and CD9 when the two cell lines were incubated with different level Elemene, Rusults: we saw that the CD42a,TSP and CD9 expression adqulis on cell surface by confocal microscopy. By flow cytometer,we saw that the positive cell numbers and mean fluorescence intensity of CD9 on PG cell are lower than on PAa cell, but the positive cell numbers and mean fluorescence intensity of TSP and CD42 a are all higher than on PAa cell. From general tendency,Elemene increase the expression of CD9 and decreased the expression of CD42a and TSP on PG and PAa cells. Conclusion: The results confimaed the metastasis capability of PG is higher than PAa, and it is one ofthe possible molecule mechanisms of Elemene inhibiting lung cancer metastasis.
