11β-羟类固醇脱氢酶在人肠癌中的表达 被引量:1

摘要 目的:观察11β-羟类固醇脱氢酶(11β-HSD)在肠癌中的表达。方法:12例确诊肠腺癌患者,术后立即取肿瘤组织和癌旁正常组织标本,采用半定量RT-PCR、Western-blotting等技术,以自身配对,研究11β-类固醇脱氢酶在肠癌中表达。结果:11β-羟类固醇脱氢酶(11β-HSD)两种同工酶在肠腺癌中表达不相同,在肿瘤组织中11β-HSD2表达明显减少,11β-HSD1在肿瘤与正常组织中表达无明显差异。结论:11β-HSD2可能与肠癌变有关。 To observe the expression of 11 beta - hydroxysteroid dehydrogenase types 1 and 2 in colorectal cancer. Methods: Neoplastic and autologous nonneoplastic tissue were collected from 12 patients with colorectal cancer. The mRNA expression ol 1 J3 - HSD1 and 11 J3 - HSD2 was detected by reverse transcription polymerase chain reaction (RT- PCR) . The protein expression of 11β- HSD1 and 11β- HSIY2 were detected by Western- blot assay. Results: Both isoforms of 11 J3 -HSD expressed in the colon adenocarcinoma, but their expression was not identical in neoplastic and non - neoplastic tissue. There was a significant decrease of 11β-HSD2 mRNA abun- dance and protein abundance in neoplastic tissue. 1118 - HSD1 protein and mRNA abundance was not significantly different from that in control tissue. Conclusion: Neoplastic transformati on is associated with decreasing of 11 11β- HSD2 mRNA and protein.
