摘要
目的观察熊果酸(UA)对胃癌细胞株MGC-803的抑制增殖作用。方法培养胃癌细胞株MGC-803,以MTT比色法及生长曲线测定UA对MGC-803抑制增殖的作用。免疫印记法测定p-ERK1/2、Cyclin D1、p21waf/cip1表达。结果MTT实验及生长曲线均显示UA明显抑制MGC-803细胞增殖,免疫印记法显示UA抑制p-ERK1/2、Cyclin D1表达,同时上调p21waf/cip1表达。结论熊果酸可以抑制MGC-803增殖,机制与影响p-ERK1/2及下游Cyclin D1、p21waf/cip1表达有关。
[ Objective ] To observe the effects of Ursolic acid on the proliferation of human gastric carcinoma MGC- 803 cell line. [Methods] The effect of Ursolic acid on the proliferation of MGC-803 cells wat- observed by MTT assay and growth curve. The expression of p-ERK1/2, Cyclin D1, p21^waf/cipl was analyzed by Western blot. [ Results ] Ursolic acid could depress the proliferation of MGC-803 cell line. The expressions of p-ERK1/2 and Cyclin D1 were decreased, while p21^waf/cipl expression was elevated by different concentrations of Ursolic acid, [ Conclusion ] Ursolic acid can depress the proliferation of MGC-803 cell line with inhibiting ERK signal transduction,
