摘要
以纸色谱(PC)和高压液相色谱(HPLC)对棉花中萜烯类化合物的种类和含量进行定性定量分析。纸色谱的流动相由己烷和乙酸乙酯组成,流动相的最佳分离比例为己烷:乙酸乙酯=1∶1。利用纸色谱对棉花的提取液定性分析发现:提取液被分离为5个斑点,由下向上各个点的Rf值依次为0.46,0.65,0.76,0.82和0.96。提取液双向纸色谱发现,单向色谱中的第3个点(中间点)又被分离成3个斑点。高压液相色谱对棉花中萜烯类化合物进行分离获得良好的分离效果,并可准确的测定各种成份的含量。在本试验条件下,半棉酚酮(HGQ)、棉酚(G)和杀实夜蛾素H1~H4的保留时间分别是3.90min,10.44min,12.99min,15.0min,14.0min和11.4min。此外,还利用HPLC法对棉花四个品种中萜烯类化合物的含量进行了测定。
Quantitative and qualitative analyses were conducted by paper chromatography (PC) and high performance liquid chromatography (HPLC) methods on the kinds and content of terpenoid in cotton varieties. The mobile phase of PC consisted of hexane and ethyl acetate and their optimum ratio separated was l to l. Five spots were separated from the extraction of cotton sample by PC, the sequential R f value of the compounds from bottom to top on the paper was 0.46, 0.56, 0.76, 0.82 and 0.96, respectively. Three spots were separated from the 3rd spot of this sequence by double direction PC. The kinds and contents of terpenoids in cotton were also analyzed by HPLC and reasonable results were obtained. Under the given conditions, the separating preserving time of hemigossypolone, gossypol and Heliothis from H 1 to H 4 was 3.90 min, 10.44min, 12.99min, 15.0min, 14.0min and 11.4min, respectively. In addition, the content of terpenoid in 4 cotton varieties was determined by HPLC.
