A simplified micro photochemical induced thrombogenesis model is recently developed. Microscope with video camera and recorder were applied for monitoring mesentery microcirculation of Wistar rats (80 ̄100g) . According to the design of Prof. Liao FL, the microscopic light(halogen bulb, 100W) was localized by a specially made aperture to illuminate and to irradiate a selected single venule with the venule diameter about 40μm and blood velocity within 0.8 ̄1.1mm/s . In the meantime, the photochemical thrombogenesis was induced by irradiation at the level of about 10 8lx incorporated with rose bengal (125mg/kg) injection. Duration between dye injection and irradiation is recommended among 5 ̄25min . Platelet thrombogenesis appeared, gradually got thick, and eventually blocked the single venule, vessel blockage time(VBT) were dye dose, light intensity dependent. The results showed that VBT was controlled at 460±123s and Action know drug, ligustrazine, effectively prolonged VBT to 676±167s ( P <0.001) . It indicated that this simplified method can be used for pharmacological application.
