摘要
目的探讨苯和环磷酰胺联合建立小鼠再生障碍性贫血模型的方法。方法按2.0mL/kg于昆明种雄性小鼠皮下注射苯油混合液,每天1次,隔日给药;并以环磷酰胺溶液(50mg/kg)腹腔注射,每日1次共7次。于给药前及后观察小鼠大体状况,外周血和骨髓液中细胞形态及数量变化,并进行小鼠肝、脾、胫骨的组织病理学观察。结果与对照组相比该方法诱导的小鼠外周血白细胞,红细胞,血小板,网织红细胞及骨髓有核细胞计数均显著减少,分别下降42.4%(P<0.O1),18.4%(P<0.05),15.4%(P<0.05),23.5%(P<0.05),24.4%(P<0.05),骨髓象及骨髓病理观察显示骨髓正常造血细胞减少,脂肪细胞等非造血细胞增加,综合以上结果提示本实验利用苯和环磷酰胺联合建立小鼠再障模型成功。结论利用苯与环磷酰胺联合诱导小鼠造血功能障碍结果稳定,发生率高,方法简单易行,各项指标与人的再生障碍性贫血相似,初步建立了苯与环磷酰胺诱发的小鼠再生障碍性贫血模型的实验方案。
Objective To establish a model of aplasfic anemia induced by benzene and cyclophosphamide in mice. Methods Kunming male mice were injected with benzene+corn oil(2.0ml/kg) once every two days and with cyclophosphamide(50mg/kg) once a day for a week.The parameters tested were as follows:general condition,peripheral blood smear,bone marrow smear,bone marrow nucleated cell count,and histopathology of liver,spleen and tibia. Results Compared with control group,experimental group mice showed the following results: leukocyte(42.4%,P〈0.01),erythrocyte(18.4%,P〈0.05),platelet(15.4%,P〈0.05),reticu-locyte(23.5%,P〈0.05) and bone marrow nucleated cell count 24.4%(P〈0.05) were decreased remarkably.The bone marrow smears and the patho-morphological examinations showed that hematopoietic cells were decreased with the increase of the non-hematopoietic cells,such as fat cells. These parameters indicated that the model was set up successfully. Conclusions An experimental aplastic anemia model in mice is established simply and easily with a high succsess rate and the results are stable.As a result,an experimental aplastic anemia model induced by benzene and Cyclophosphamide in mice is preliminarily set up.
