鼻息肉组织辅助性T细胞亚群及相关转录因子表达 被引量:3

摘要 目的探讨合并与不合并变应性鼻炎(allergic rhinitis,AR)的鼻息肉组织在免疫应答方面的差异。方法标本取自16例不合并 AR 的鼻息肉患者及16例合并 AR 的鼻息肉患者。新鲜鼻息肉标本制备细胞悬液,流式细胞术测定辅助性 T 细胞(helper T cell,TH)1和 TH2的百分率。液氮冷冻鼻息肉标本实时荧光定量 PCR 测定 TH 分化相关转录因子 GATA 结合蛋白3(GATA-binding protein 3,GATA-3)及 T 细胞表达 T-box(T-box expressed in T cells,T-bet)mRNA 的表达。结果鼻息肉组织以 TH1亚群为主;合并 AR 的鼻息肉组织TH2百分率显著高于不合并 AR 的鼻息肉组织(P<0.01);鼻息肉组织 GATA-3及 T-bet mRNA 表达显著高于下鼻甲组织(P<0.01);合并 AR 的鼻息肉组织 GATA-3 mRNA 表达显著高于不合并 AR 的鼻息肉组织(P<0.01)。结论合并与不合并 AR 的鼻息肉组织在免疫应答方面具有不同特征,细胞免疫参与了不合并 AR 的鼻息肉的发生过程,细胞免疫和体液免疫均参与了合并 AR 的鼻息肉的发生过程。(中国眼耳鼻喉科杂志,2007,7:285-287) Purpose To investigate the difference in immunological pattern of nasal polyps in patients with and without allergic rhinitis (AR). Methods Sixteen cases of polyps without AR and sixteen with AR were involved in this study. The fresh samples were prepared into cell suspension for flow cytometric analysis. The liquid nitrogen stored tissues were analyzed with real-time quantitative PCR for the expression of GATA-binding protein ( GATA-3 ) and T-box ex pressed in T cells (T-bet) mRNA. Results TH1 subset was predominant in all the nasal polyps. The mean percentage of TH2 in nasal polyps with AR was significantly higher than that in the nasal polyps without AR. The nasal polyps demonstrated higher expression of GATA-3 and T-bet mRNA than that in the inferior turbinate tissue. The nasal polyps with AR showed higher levels in GATA-3 mRNA expression than that in the nasal polyps without AR. Conclusions The nasal polyps with and without AR are different in immunological patterns. Cytoimmunity participate in the pathogenesis of nasal polyps without AR. Cytoimmunity and humoral immunity are both involved in the pathogenesis of nasal polyps with AR. ( Chin J Ophthalmol and Otorhinolaryngo1,2007,7:285-287)
