疏肝养胃通脉冲剂对慢性萎缩性胃炎患者DNA错配基因hMSH_2蛋白表达的影响

Effect of Shugan Yangwei Tongmai Granules on Expression of DNA Mismatching Gene hMSH_2 Protein in Chronic Atrophic Gastritis Patients

目的观察疏肝养胃通脉冲剂对慢性萎缩性胃炎(CAG)患者DNA错配基因hMSH2蛋白表达的影响。方法应用免疫组化S-P方法,对174例不同病理亚型CAG患者进行治疗前后hMSH2蛋白表达检测。结果治疗组随着萎缩加重、异型加大和Hp感染程度加深,治疗后下调hMSH2蛋白过表达加大(P<0.05);而对照组治疗前后变化不显著。在化生中,治疗组随肠上皮和假幽门腺化生加重,细胞核hMSH2蛋白过表达下调渐小,呈负相关性,即单纯组最显著(P<0.01),细胞质以中度最显著(P<0.01),重度组虽有下调,但治疗前后比较相关性不显著(P>0.05);对照组细胞核和细胞质hMSH2蛋白过表达有一定的下调,但同组间比较相关性不显著(P>0.05)。结论疏肝养胃通脉冲剂能抑杀Hp,对CAG患者特别是伴异型增生者能下调hMSH2蛋白过表达,对治疗CAG和预防其癌变有明显效果。

Objective To observe the effect of Shugan Yangwei Tongmai Granules （Granule for Soothing Liver, Nourishing Stomach and Removing Obstruction） on expression of DNA mismatching gene hMSH2 protein in chronic atrophic gastritis （CAG） patients. Methods The S-P immunohistochemical method was used to test the hMSH2 protein expression of 174 atypical CAG cases. Results In the treatment group, with the atrophy aggravation, heterotype increase and Hp infection intensify, the decrease of over-expression of hMSH2 protein was increased after treatment （P〈0.05）. In metaplasia, the intestinal and pseudopyloric gland metaplasia was aggravated, the decrease of over-expression of hMSH2 protein of nucleolus showed a negative correlation, i.e. that of the simple group was significant （P〈0.01）, that of the cytoplasm in the medium group was significant （P〈0. 01）, and in the severe group it was decreased, but not significantly （P〉0.05）. In the control group, the over-expression of hMSH2 protein of nucleolus and cytoplasm was decreased to a certain degree, but the in-group comparison correlation was not significant （P〉0. 05）. Conclusion The Chinese herbal medicine Shugan Yangwei Tongmai Granules could inhibit and kill Hp and decrease the over-expression of hMSH2 protein of CAG, especially for those accompanied with atypical hyperplasia, being effective for treating CAG and prevent its carcinogenesis.
