摘要
目的:探讨三氮唑席夫碱衍生物对人肝癌细胞SMMC-7721的诱导分化作用.方法:用不同剂量的3-吡啶-3-基-4-[(4-羟基-3-甲氧基-苯亚甲基)氨基]-5-甲硫基-1,2,4-三唑(LH-37)与SMMC-7721细胞共同培养,以MTT法检测细胞增殖和LH-37对细胞增殖的抑制作用;镜下观察细胞形态改变;采用RT-PCR方法检测细胞甲胎蛋白mRNA表达,金氏法检测细胞碱性磷酸酶活性的变化.结果:LH-37在1×10^-5~1×10^-3mol/L的浓度范围均能抑制细胞增殖,且抑制率随浓度的增加而增高(P〈0.05或P〈0.01),细胞形态和结构向正常肝细胞方向逆转,1×10^-5mol/L和1×10^-6mol/L的LH-37均可使细胞甲胎蛋白mRNA表达量明显下降,碱性磷酸酶活性明显升高.结论:三氮唑席夫碱衍生物对人肝癌SMMC-7721细胞具有抗增殖和促分化作用.
AIM: To study the effect of 3-pyridin-3-yl-4-[ (4- hydroxy-3-methoxy-benzylidene ) amino ] -5-methylsulfanyl-4H- [ 1, 2,4] triazole (LH-37) on the induction of differentiation of hepa- tocarcinoma SMMC-7721 cells. METHODS: SMMC-7721 cells were cultured in RPMI-1640 and treated by LH-37 at different doses. The proliferation of the cells was examined by MTT assay. Cell morphology was observed by light microscopy. The reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the changes of alpha fetoprotein (α-FP) mRNA. The spe- cific activities of alkaline phosphatase (ALP) were assayed by ALP kit based on King's method. RESULTS: The cell proliferation was inhibited by LHo37 at 1 × 10^ -5- 1 × 10^ -3 moL/L in dose dependent manners. 1 ×10^-5 mot/L LH-37 induced the morpho- logical normality of SMMC-7721 cells. After treated with 1 × 10 ^-5 mot/L or 1 × 10^-6 mot/L LH-37, the mRNA expression of α-FP was significantly reduced and the activities of ALP were remar- kably declined ( P 〈0.01 ). CONCLUSION: LH-37 may inhibit proliferation and induce differentiation of human hepatocarcinoma SMMC-7221 cells.
