独占春种子非共生萌发和低温离体保存 被引量:3

摘要 独占春(Cymbidium eburneum Lindley)成熟种子无菌播种诱导原球茎,并以原球茎进行低温离体保存。结果表明:在培养基花宝1号3g/L+LH(水解乳蛋白)1g/L+BA(细胞分裂素)0.5 mg/L+NAA(生长素)0.1 mg/L+CM (椰子汁)100 ml/L+AC 2 g/L+蔗糖25 g/L中种子萌发出原球茎;原球茎在培养基1/2 MS+LH 0.5 g/L+BA 0.5 mg/L+NAA 0.1mg/L+AC 2g/L+蔗糖20g/L中于6℃黑暗条件下连续保存了20个月,成活率达85%,并能在恢复培养基MS+BA 3~5 mg/L+NAA 0.2 mg/L+AC 1g/L+CM 100 ml/L+蔗糖25 g/L中进行恢复增殖培养。 Seeds of Cymbidium eburneum Lindley were sown aseptically to induce protocorms. The protocorms were preserved in vitro at low temperature. The results showed the seeds were germinated into protocorms on the culture medium of Hyponex-1 3 g/L + LH 1 g/L + BA 0.5 mg/L + NAA 0.1 mg/L + CM 100 ml/L + AC 2 g/L+ sugar 25g/L. And the protocorms were conserved for 20 months at 6℃ in dark without subculture on the culture of 1/2MS + BA 0.5 mg/L + NAA 0.1 mg/L + AC 2 g/L + LH 0.5 g/L + sugar 20 g/L, and their survival rate was upto 85%. They started to multiply when placed on the culture medium of MS + BA 3-5 .mg/L + NAA 0.2 mg/L + AC 1 g/L + CM 100 ml/L + sugar 25 g/L.
