高效液相色谱法测定保健食品中天麻素含量

Determination of gastrodin in health food by HPLC

目的：建立保健食品中天麻素的高效液相色谱测定方法。方法：样品以甲醇提取，中性氧化铝柱净化，ZORBAX Eclipse PlusC18（4．6×250mm）柱分离，流动相为甲醇-磷酸盐溶液-水（7：3：90），流速：1．0ml／min，紫外检测器检测，波长：223nm。结果：天麻素在5—25μg/ml有良好的线性关系，相关系数为0．9995。检出限为0．04μg/ml，最低检出浓度为0．08mg／kg。样品测定的精密度为2．8％-3．0％；样品加标回收率为95．6％-96．6％，平均回收率为96．0％。结论：本方法简便快速、检测灵敏度高，适用于保健食品中天麻素的测定。

Objective：To develop a HPLC method for the determination of gastrodin in health food. Methods：Gastrodin in health food was extracted by methanol and purified through a column filled with aluminium oxide（neutral） ,the separation was performed on a ZORBAX Eclipse Plus C18 column（4. 6 ×250 mm,5 μm）with the mobile phase consisting of methanol - phosphate buffer solution - water（7：3：90,v/v） with a flow rate of 1.0 ml/min,the determination was made by a U. V. detector at 223 nm wavelength. Results：The method offered good linearity of5.0 - 25 μg/ml,relativity coefficient was 0. 9999 ,the detection limit was 0. 04 μg/ml,the average recovery was 96.0%. the precision of method was from 0. 9% - 1.2% （ a =6）. Conclusion：The developod HPLC method is accurate and precision, and can be used for the determination of gastrodin in health food.
