摘要
目的:建立保健食品中天麻素的高效液相色谱测定方法。方法:样品以甲醇提取,中性氧化铝柱净化,ZORBAX Eclipse PlusC18(4.6×250mm)柱分离,流动相为甲醇-磷酸盐溶液-水(7:3:90),流速:1.0ml/min,紫外检测器检测,波长:223nm。结果:天麻素在5—25μg/ml有良好的线性关系,相关系数为0.9995。检出限为0.04μg/ml,最低检出浓度为0.08mg/kg。样品测定的精密度为2.8%-3.0%;样品加标回收率为95.6%-96.6%,平均回收率为96.0%。结论:本方法简便快速、检测灵敏度高,适用于保健食品中天麻素的测定。
Objective:To develop a HPLC method for the determination of gastrodin in health food. Methods:Gastrodin in health food was extracted by methanol and purified through a column filled with aluminium oxide(neutral) ,the separation was performed on a ZORBAX Eclipse Plus C18 column(4. 6 ×250 mm,5 μm)with the mobile phase consisting of methanol - phosphate buffer solution - water(7:3:90,v/v) with a flow rate of 1.0 ml/min,the determination was made by a U. V. detector at 223 nm wavelength. Results:The method offered good linearity of5.0 - 25 μg/ml,relativity coefficient was 0. 9999 ,the detection limit was 0. 04 μg/ml,the average recovery was 96.0%. the precision of method was from 0. 9% - 1.2% ( a =6). Conclusion:The developod HPLC method is accurate and precision, and can be used for the determination of gastrodin in health food.
