摘要
目的检测Caspase家族中Caspase-3在膀胱移形细胞癌(BTCC)组织及正常膀胱组织中的表达,探讨Caspase-3的表达在膀胱癌发生发展中的意义。方法采用免疫组织化学和实时荧光定量逆转录-聚合酶链反应(RT-QPCR)方法对30例膀胱癌和15例正常膀胱组织中Caspase-3的表达进行检测。结果免疫染色标本中,在正常膀胱组织和膀胱癌中Caspase-3的阳性表达率分别为80%和30%,两者差异有统计学意义(P<0.05)。Caspase-3在膀胱癌组织中的△△CT值是正常膀胱组织的1.9274(1.1248~2.5413)倍,但病理分级之间和临床分期之间均差异无统计学意义。结论Caspase-3表达下调导致细胞凋亡减少,可能在膀胱癌的发生发展过程中起重要作用。
Objective To study the expression of Caspase-3 in bladder transitional cell carcinoma (BTCC) tissues and normal bladder tissues and explore the clinical significance of Caspase-3 in the development of BTCC. Methods In 30 bladder cancer tissues and 15 normal bladder tissues ,the expression of Caspase-3 was detected by using immunohistochemistry and real-time quantitative reverse transcription polymerase chain reaction (RT-QPCR). Results The positive rate of Caspase-3 immune-staining specimens was 80% ( 12/15 ) and 30% (9/30) in normal bladder tissues and BTCC tissues, respectively (P 〈 0.05 ). The A ACT value of Caspase-3 in bladder cancer tissues was 1. 9274 ( 1. 1248-2. 5413 ) times of that in normal bladder tissues. There was no significant difference in the expression of Caspase-3 mRNA among tumor pathological grades or clinical stages. Conclusion Downexpression of Caspase-3 results in the reduction of tumor cell apoptosis ,which may play an important role in development and progression of bladder cancer
