人精子运动功能与腹腔液中干扰素-γ含量的关系 被引量:1

Correlation of sperm motility with the levels of IFN-γin peritoneal fluid

摘要 目的观察子宫内膜异位症(EMs)患者腹腔液干扰素(IFN)-γ体外对精子运动功能的影响。方法用双抗体夹心法,检测30例EMs不孕妇女,30例其他病因所致不孕妇女和20例生育力正常妇女腹腔液中IFN-γ含量,用精液自动分析系统检测与腹腔液共同孵育后的正常精液中精子活动力参数,荧光显微镜观察顶体反应发生率。结果EMs组腹腔液IFN-γ含量(250.3±102.6)ng/L高于其他2组(P<0.01);EMs组腹腔液与正常精液共同孵育后,精子活动力显著降低(P<0.01),并与IFN-γ含量呈负相关(r=-0.958,P<0.01)。精子与EMs组腹腔液孵育16 h后顶体反应发生率低于正常对照组(P<0.01)。结论EMs患者腹腔液IFN-γ对精子活动的抑制可能是导致不孕因素。 Objective To evaluate the influence of IFN-γ in peritoneal fluid of patients with endometriosis on human sperm motility in vitro, and explore the pathogenesis of infertility associated with EMS. Methods The level of IFN-γ in peritoneal fluid of 30 EMS women was measured by enzyme linked immunosorbent assay, and compared with that of 30 non-EMS women with infertility and 20 normal women. The parameters of sperm motility were tested by sperm automatic analysis system after sperm was incubated with peritoneal fluid. Sperm acrosome reaction was observed by the fluorescence microscope. Results IFN-γ contents in EMS group (250.3 ± 102.6) ng/L were significantly higher than those in other two groups ( P 〈 0.01 ). Sperm motility was suppressed after sperm was incubated with peritoneal fluid in EMS group as compared with that in other two groups (r = -0.958 ,P 〈 0.01 ), while sperm motility level was negatively-correlated with contents of IFN-γ in peritoneal fluid of EMS group. Sperm acrosome reaction in EMS group was reduced significantly in comparison with the control 16 h after incubation with peritoneal fluid ( P 〈 0.01 ). Conclusion It is likely that reduction of sperm motility by IFN-γ in peritoneal fluid in patients with EMS may play a role in the pathogenesis of infertility associated with EMS.
