摘要 目的研制抗金环蛇毒血清,以解决被金环蛇咬伤后的救治问题。方法以金环蛇毒为免疫原免疫马获得免疫血浆,经盐析制备IgG,IgG再经酶解、疏水柱纯化后得到F(ab′)2活性片段,并以免疫扩散、ELISA测定其抗体特异性,以小鼠试验法测定其中和金环蛇毒能力。结果得到的抗血清F(ab′)2冻干品纯度达90%以上。结论研制出了特异性强、效价高的抗金环蛇毒血清。 Objective To prepare the Bungarus fasciatush antivenin to cure the snakebite which caused by the title snake. Methods The IgG was firstly prepared by ammonium sulfate precipitation from horse plasma which immunificated by Bungarus fasciatush venom, then subjected to pepsin-digesting and hydrophobic chromatography, the F(ab′)2 active fragment was obtained. The specificity of antivenin was traced through ELISA and the immunodiffusion, as well as protective test was performed on mice in vitro. Results The purity of F (ab′)2 active fragment was over 90%. Conclusion The study led to the preparation of Bungarus fasciatush antivenin with high speciality and high potency.
