摘要
目的:探讨醛固酮受体拮抗剂安体舒通对大鼠单侧输尿管梗阻(UUO)模型肾间质纤维化的抗氧化保护作用。方法:Wistar大鼠行左侧输尿管结扎术,分为UUO模型组(n=11)和安体舒通治疗组(n=12),同时设假手术对照组(n=7)。术后第14天处死各组大鼠,进行HE和Masson染色,观察肾脏病理变化;比色法测定肾组织丙二醛(MDA)和超氧化物歧化酶(SOD)含量;放免法测定血浆和组织醛固酮(ALD)含量。结果:UUO模型组与假手术组比较,肾脏病理改变加重,肾组织MDA含量明显升高,SOD含量显著下降,血浆和组织ALD含量明显升高(P<0.05)。安体舒通组与UUO组大鼠比较,肾间质纤维化显著减轻,肾组织MDA含量明显降低,SOD含量显著升高,血浆和组织ALD含量下降(P<0.05)。结论:安体舒通可能减少单侧输尿管梗阻肾组织脂质过氧化物的产生,同时增加抗氧化酶的含量,从而改善UUO大鼠氧化应激所致的肾间质纤维化。
Objective:To investigate the antioxiative effects of spironolactone ,an aldosterone receptor blockade,on kidney of rats with unilateral ureteral obstruction (UUO). Methods:Thirty wistar rats underwent UUO and then received vehicle (n = 11) or spironolactone(50mg·kg^-1·d^-1 by daily gastric gavage,n = 12) during the 15 days from the day before surgery. Additional 7 rats were sham operated. All the rats were killed at 14 days after surgery. Histological changes were observed by HE and Masson staining. Malondialdehyde (MDA), superoxide dismutase (SOD) as well as aldosterone (ALD) content were measured. Results: UUO induced a significant increase in MDA and ALD content but a marked decrease in SOD content and severe morphology changes. However, spironolactone ameliorated what had mentioned. Conclusions :Spironolactone can suppress oxidative stress and alleviate renal in rats with UUO by reducing lipid peroxidation production and improving antioxidative enzyme contents.
