摘要
目的探讨变应性鼻炎(allergic rhinitis,AR)患者外周血单个核细胞(peripheral bloodmononuclear cell,PBMC)内两种皮质类固醇(corticosteroids,CS)受体亚型——CS-α和 CS-β表达水平与类固醇治疗效应之间的关系。方法采用半定量 RT-PCR 方法检测类固醇不同效应 AR 患者和健康对照组 PBMC 的 CS-α、CS-β mRNA,采用免疫细胞化学方法检测 CS-α和 CS-β蛋白的表达。结果①类固醇敏感组、类固醇抵抗组及健康人体内均有 CS-α、β mRNA 的表达,但以 CS-α为主,其 CS-α/GAPDH(葡萄糖甘油醛脱氢酶)mRNA 的比值(±s,下同)分别为1.15±0.75,1.63±0.78,1.27±0.51;②类固醇抵抗组 CS-β mRNA 表达明显高于类固醇敏感组和健康对照(P<0.05),3组 CS-β/GAPDH mRNA 的比值分别为1.42±0.73,0.82±0.59,0.80±0.68;③免疫细胞化学结果显示,类固醇抵抗组 PBMC 的 CS-β表达阳性细胞比率明显高于类固醇敏感组及健康对照组(P<0.01),分别为28.8%±9.9%,5.9%±3.2%,5.5%±6.8%。结论变应性鼻炎患者类固醇抵抗可能与 CS-β的表达亢进有关。
Objective To clarify the relationship between the expression of α- and β-isoform of corticosteroid receptors(CS) in peripheral blood mononuclear cell(PBMC) and response to corticosteroid in patients with allergic rhinitis (AR). Methods Semi-quantitative RT-PCR was used to detect the expression of CS-α、β in PBMC in patients with AR and to observe the different responses to corticosteroid in controls. Inmaunocytochemical assay was used to detect the expression of protein of CS-α- CS and β. Results ① The expression of CS-α mRNA was detected in the sensitive group and the resistant group of patients with AR and the controls with CS-α/GAPDH mRNA (x^-±s) 1.15±0.75,1.63±0.78,1.27±0.51 respectively. ② The expression of CS-13 mRNA in PBMC in the resistant group of patients with AR was significantly higher than that in the sensitive group and the controls( P 〈 0. 05 ), with CS-β/GAPDH mRNA 1.42±0.73,0.82±0.59,0.80±0.68 respectively. ③ The number of CS-β-positive PBMC in the resistant group was significantly higher than that in the sensitive group and the controls (P 〈0.01 ), with the number of CS-β-positive PBMC 28.8%±9.9%,5.9%±3.2%,5.5%±6.8% respectively. Conckysuibs It is shown that the excessive expression of CS-β may serve as a novel predictor of corticosteroid resistance in patients with AR.
