摘要
目的:为了确定沈阳地区新分离汉坦病毒的基因分型及其M片段G1、G2区和S片段的序列特征,对分离株进行了系统进化分析。方法:通过免疫荧光法筛查阳性标本;组织细胞培养法分离汉坦病毒;用RT-PCR法分别扩增分离株的M片段G1、G2区和S片段,测序后应用DNAstar软件同已知标准株进行碱基配对和比较分析,种系发生采用PHYLIP软件包进行分析。结果:通过系统进化树的分析表明,分离株与HTN型株同源性较高,同源性达到95.0%~97.0%。结论:沈阳地区汉坦病毒分离株与BAO 10、BAO 14、JIANG 13、BAO 9、Q33处于同一分支,属于同一HTN型。
Objective:To determine genetic type of the new isolated hantavirus genotype found in Shenyang and the sequence characteristic of G1,G2 fragments of M segment and S segment of the virus,as well as to do a systematic evolution analysis of the isolated strain.Methods:The positive specimen were screened by the means of immunity fluorescence and hantavirus isolated through cell culture.The G1,G2 fragments of M segment and S segment of the isolated strain were amplified respectively by means of RT-PCR in order to do base pairing with standard strain and comparison analysis through software of DNA Star.Phylogeny was analyzed through PHYLIP software.Results:The isolated strain of hantavirus and HTN showed a higher homology,going up to 95.0%~97.0%.Conclusion:The isolated strain of hantavirus in Shenyang is on the same branch with BAO 10,BAO 14,JIANG 13,BAO 9,Q 33 and all of them belong to HTN.
