摘要
目的探讨内皮素-1(endothelin-1,ET-1)对人视网膜色素上皮细胞(retinalpig-mentepithelium,RPE)黏附作用的影响。方法用RPE黏附实验和放射免疫测定法检测不同浓度ET-1对RPE黏附的影响和6-Keto-PGFα1的浓度变化。结果10-11~10-9mol.L-1ET-1对RPE细胞黏附力呈剂量依赖性增加,而RPE细胞黏附力随着ET-1浓度的进一步增加(10-9~10-7mol.L-1)呈剂量依赖性下降(F=6.352,P<0.01);黏附实验上清液中6-Keto-PGFα1含量随着ET-1浓度增加呈浓度依赖性增加(F=33.929,P<0.01)。结论ET-1对RPE有促进黏附作用,ET-1刺激后6-Keto-PGFα1含量上升,与RPE细胞对ET-1作出的反应有关,并抑制ET-1作用。
Objective To investigate the adherent effects of endothelin-1 (ET-1) on the human retinal pigment epithelial (hRPE) cells. Methods RPE attachment test and radioimmunoassay were used to detect adherent effects on hRPE cells and content change of 6-Keto-PGFα1 in different concentrations of ET-1. Results The RPE cell adhesion increased after stimulated by ET-1 doses from 10^-11·mol·L^-1 to 10^-9 mol·L^-1 in a dose-dependent manner and decreased from 10^-9mol·L^-1 to 10^-7 mol·L^-1(F=6.352,P〈0.01).The contents of 6-Keto-PGFα1 in RPE cell conditioned medium increased in a dose-dependent manner after stimulated byflve ET-1 doses from 10^-11mol·L^-1 to 10^-7 mol·L^-1 (F=33.929,P〈 0.01 ). Conclusion ET-1 stimulates the adhesion of RPE cells. The increase of 6-Keto-PGFα1 in the cell-conditioned medium may be respond to ET-1, and inhibits the effects of ET-1.
