摘要
为在短期内了解β-葡聚糖酶的储存稳定性,用加速实验估算了β-葡聚糖酶在不同温度下的储存寿命。选择的实验温度为28、36.2、45℃。将盛有β-葡聚糖酶的样品瓶放入不同温度的水浴中,不同时间取样并测定β-葡聚糖酶活性。实验数据的统计处理结果得出:β-葡聚糖酶在45、36.2℃和28℃活性损失10%所需时间为1.36、4.37和20.09d;活性损失50%所需时间为8.5、25.47、104.9d。由寿命估算曲线估算出β-葡聚糖酶在25、20、15、4℃和0℃时,性损失10%需要的时间分别为30.7、67.0、145.9d,2.21年,4.14年;活性损失50%所需时间为154.0、316.0、648、3154d。从加速实验的结果可以看出β-葡聚糖酶的稳定性可以满足储存的需要。加速储存实验可以在较短时间内估算出β-葡聚糖酶储存寿命。
In order to study rapidly on the storage stability of β-glucanase, the accelerated storage test was carried out to estimate the shelf life of β-glucanase at different temperature. The samples was kept in the water bath at 28℃, 36.2℃, 45℃ and its activity was determined at different time. The results of statistical treatment of data indicate that the activity of β-glucanase decreases by 10% after 1.36d, 4.37d, 20.09d, 30.7d, 67.0d, 145.9d, 2.21years, 4.14 years and decreased by 50% after 8.5d, 25.47d, 104.9d, 154.0d, 316.0d, 648d, 3154d when it was kept at 45℃, 36.2℃, 28℃, 25℃, 20℃, 15℃, 4℃ and 0℃, respectively. The results indicate that the stability of β-glucanase is competent for the storage, and the accelerated storage test can estimate the shelf life of β-glucanase.
