摘要
以辣椒细胞质雄性不育系21A及其同核异质保持系21B为试验材料,比较分析两系雄配子发育过程中酯酶(EST)、谷氨酸脱氢酶(GDH)和苹果酸脱氢酶(MDH)同工酶的表达特征.结果表明:幼叶和花蕾的EST同工酶酶谱在谱带数目和酶带强弱上存在时空表达差异,并且随着雄配子发育的进行,保持系21B从中花蕾至特大花蕾比不育系21A多1条清晰的谱带(EST3e),其差异表达发生在细胞学上观察到的败育时期之前;在GDH同工酶中,保持系21B从大花蕾至特大花蕾比不育系多6条谱带(GDH1和GDH1/2),酶谱差异表达时期与细胞学上观察到的败育时期一致;而在MDH同工酶中,不育系21A和保持系21B的幼叶和各级花蕾的酶谱在谱带数目和谱带强弱上均没有明显差异.
21A and 21g,which were the cytoplasmic male sterility line and its maintainer of Capsicum annuurn, were used to comparatively analyze the expression of esterase (EST), glutamate dehydrogenase (GDH) and malate dehydrogenase (MDH) isozyme in young leaves and flower buds at different stages of flower development. The results showed that the difference of the number and intensity of the bands in zy- mograms of EST were observed between young leaves and flower buds of pepper,presenting spatiotemporal expression patterns. With the male gamete developing, the maintainer line 21B had one more band (EST3e) than that of the cytoplasmic male sterility line 21A. The differences expressions of EST zymogram between 21A and 21B occurred before cytological abortion could be observed. For GDH zymogram, the maintainer line 21B had six more bands (GDH1 and GDH1/2 ) than that of the cytoplasmic male sterility line 21A from the large flower buds stage to the extremely large flower buds stage. The different expressions of GDH zymogram between 21A and 21B occurred at the same stage of cytological abortion. No any difference of the number and intensity of the bands in zymograms of MDH were observed in young leaves and flower buds at different stages of flower development between 21A and 21B.
