摘要
本文应用碱性单个细胞微量凝胶电泳测试技术对1,1-二氯乙烯致大鼠肝细胞DNA损伤进行了研究,结果显示:1,1-二氯乙烯可引起肝实质性细胞DNA链的断裂,而且发现肝实质性细胞较非实质性细胞敏感。结果还表明,单个细胞微量凝胶电泳技术是一项非常敏感的测试哺乳动物细胞DNA损伤的方法。本文还就1,1-二氯乙烯致肝细胞DNA损伤的可能机制进行了讨论。
Vinylidene chloride was assayed using microgel single cell electrophoresis(MGE) technique for its ability to induce genotoxic effect in the freshly isolated rat hepatocytes in vitro. The results showed that vinylidene chloride clearly produced a dose dependent induction of DNA strand breaks in parenchymal cells of rat liver at doses equal to or exceeding 10μg/ml. Different cell susceptibility to induced DNA damage was observed. Our results also indicated that the microgel single cell electrophoresis assay was more sensitive and revealed the genotoxic potential of vinylidene chloide at lower doses than the other mutation assay.
