摘要
在淡色库蚊的乙酰胆碱酯酶ace1基因上发现了1个可变剪接,与实验室敏感株1个个体的ace1基因的编码区相差了24个碱基,导致所编码的成熟蛋白在44~51氨基酸残基处缺失了8个氨基酸。经过在昆虫杆状病毒表达系统中的表达,推测这段基因的可变剪接可影响乙酰胆碱酯酶的活性。
An alternative splicing is found in ace 1 gene from a sensitive individual mosquito Cx. p. pallens. This is the first report of alternative splicing in an AChE gene from mosquitoes. The splicing site is found in 44 - 51 AA of mature protein after cleavage of the leader peptide. Two proteins which differed in size by eight amino acids were expressed by Bac-To-Bac Baculovirus Expression System. The enzyme activity data indicated that the splicing form of ACHE1 of Cx. p. pallens was less activity than the non-splicing form. A further research will be necessary to be done on the functions of the alternative splicing in ACHE1 of Cx. p. pallens, such as affinity, substrate specificity, catalytic properties, Vmax and activity regulation.
