三羟基异黄酮对人多发骨髓瘤细胞的增殖及细胞内NF-κB的影响 被引量:1

摘要 目的:研究4′,5,7三羟基异黄酮(Genistein)对人多发骨髓瘤(multiple myeloma,MM)细胞的增殖及NF-κB表达的影响。方法:体外培养人多发骨髓瘤细胞株XG1,依据剂量梯度分别给予Genistein,用噻唑蓝染色法(MTT)观察不同药物浓度的Genistein对MM细胞的增殖影响;应用10μg/mL Genistein处理XG1细胞,用免疫组化法观察Genistein处理前后细胞内NF-κB的表达情况。结果:Genistein作用XG1细胞72h,随浓度增加,细胞增殖逐渐下降,半数抑制浓度在11μg/mL左右;Genistein使NF-κB在XG1细胞内重新分布,胞核内NF-κB表达下降。结论:Genistein可能通过下调骨髓瘤细胞核中NF-κB表达来抑制骨髓瘤细胞的增殖。 OBJECTIVE: To investigate the effect of genistein on proliferation and NF -κB of multiple myeloma XG1 cells. METHODS: XG1 cells were treated with a series of concentrations of genistein. The proliferation of XG1 cells was measured with MTT assay. The expression of NF-κB was detected with immunocytochemistry after the XG1 cells was treated with 10 μg/mL Genistein, RESULTS: After exposed to genistein for 72 hours, the proliferation of XG1 was inhibited in concentration-dependent manner, and IC50 was 11 μg/mL. Genistein decreased NF-κB in XG1 cellular nucleus. CONCLUSION: Genistein can inhibit the proliferation of XG1 cells by affecting the distribution of NF-κB in cells.
