肺组织血管生成变化与肺纤维化关系探讨 被引量:9

摘要 目的观察肺纤维化大鼠肺组织血管生成状态的变化,初步探讨血管生成与肺纤维化形成的关系。方法复制博来霉素诱发肺纤维化大鼠模型,用HE染色结合因子Ⅷ相关抗原(vWf)免疫组织化学方法观察模型组和对照组肺组织血管生成状态,采用羟脯氨酸含量测定法评估肺纤维化程度。结果模型组大鼠肺组织血管生成状态自第1d开始即明显活跃,血管计数升高,第7d达高峰,肺组织胶原沉积也相继出现;随着肺纤维化加重,血管生成逐渐减少,至第21d之后低于对照组水平。结论血管生成在肺纤维化早期显著增加,可能是肺纤维化形成中的重要启动环节。 Objective To observe the changes of angiogenesis in experimental pulmonary fibrosis in rats and the relationship between them. Methods Bleomycin-induced rat model of pulmonary fibrosis was established. Angiogenesis was evaluated by HE staining and anti-von willebrand factor (anti-vWf) immunohistochemistry. Fibrosis was judged by hydroxypmline concentration assay. Results Angiogenesis in the model group was observed from day 1, climaxing on day 7, and the hydroxypmline concentration increased along with angiogenesis. As fibrosis developed angiogenesis decreased gradually which was lower than that in control group on day 21. Conclusion Angiogenesis was significant in the early stage of pulmonary fibrosis which may be an important initiating mechanism in the fibrosis process.
