摘要
目的观察亚硒酸钠和维甲酸联合作用对HL┐60细胞DNA与蛋白质合成和细胞周期的影响。方法以人早幼粒白血病细胞(HL┐60)为实验对象,利用3H┐脱氧胸苷(3H┐TdR)和3H┐亮氨酸(3H┐Leu)参入实验以及流式细胞仪进行分析。结果5.8μmol/L亚硒酸钠和0.1μmol/L维甲酸(RA)联合可显著抑制HL-60细胞的DNA和蛋白质合成,联合作用强于两者单独作用。流式细胞分析的结果表明:单独硒组的HL┐60细胞在G2+M期和S期有一定的阻滞;维甲酸的作用主要是阻滞G1期向S期移行,使G1期细胞蓄积,但该剂量单独作用较小;联合组主要表现在G1期阻滞作用要大于单独RA组的作用。
Objective:Combined effect of sodium selenite (Na 2SeO 3) and all trans retinoic acid (RA) on DNA and protein syntheses and cell cycle of HL 60 cell line was studied. Methods: Assays of 3H TdR and 3H Leu incorporation and the flow cytometric analysis were undertaken. Results: Either 5 8μmol/L Na 2SeO 3 or 0 1μmol/L RA decreased the syntheses of DNA and protein,but combination of two chemicals decreased more significantly. Na 2SeO 3 could slightly arrest HL 60 cells at G 2+M and S phases,RA increased G 1 and reduced S and G 2 cells,while Na 2SeO 3 plus RA increased G 1 more significantly Conclusions: Combined effect of Na 2SeO 3 and RA was much stronger than either of the two chemicals alone.
