摘要
目的探讨阿维A和窄谱中波紫外线(NB-UVB)对培养的正常人角质形成细胞增殖和肝素结合表皮生长因子样生长因子(HB-EGF)mRNA表达的影响。方法用1μmol/L阿维A和/或100 mJ/cm^2 NB-UVB处理正常人角质形成细胞12 h后,用MTT法、RT-PCR和实时荧光定量RT-PCR法分别检测细胞增殖和HB-EGF mRNA表达的改变。结果1μmol/L阿维A和100 mJ/cm^2 NB-UVB单独作用时,均可抑制角质形成细胞的增殖和上调HB-EGF mRNA的表达,两者联合作用时,对角质形成细胞增殖的抑制和HB-EGF mRNA的诱导作用更强,分别为19.7%和8.6倍,之后依次为1μmol/L阿维A(14.4%,7.1倍)和100 mJ/cm^2 NB-UVB(8.7%,2.5倍)。结论阿维A和/或NB-UVB处理后,可协同上调HB-EGF的表达,可能参与调节二者对角质形成细胞增殖的抑制作用。
Objective To investigate the effect of acitretin and narrow-band UVB ( NB-UVB ) on the cell proliferation and mRNA expression of heparin-binding epidermal growth factor like growth factor ( HB-EGF ) of cultured normal human keratinocytes. Methods Keratinocytes were isolated from the foreskin of young adults, and subjected to primary culture. After 3-5 passages, the keratinocytes were cultured with 1 μmol/L acitretin for 12 h and/or irradiated with 100 mJ/cm^2 NB-UVB. The proliferation potency of cells was detected by MTT colorimetric assay, and mRNA expression of HB-EGF by reverse transcription polymerase chain reaction (RT-PCR) and real-time quantitative RT-PCR. Results The treatment with acitretin and/or NB-UVB inhibited the keratinocyte growth and upregulated the mRNA expression of HB-EGF. The keratinocyte proliferation was inhibited by 14.4%, 8.7% and 19.7%, and the mRNA expression of HB-EGF was elevated by 7.1, 2.5 and 8.6 folds, respectively by 1μ mol/L acitretin, 100 mJ/cm^2 NB-UVB and the combination of them. Conclusions Acitretin and NB-UVB could synergistically upregulate the expression of HB-EGF, which may be involved in the modulation of the inhibitory effect on the proliferation of keratinocytes.
