摘要
采用不同浓度的NTG诱变剂,对南昌霉素产生菌南昌链霉菌80-5.3-116菌株的孢子进行处理。结果表明,500μg/ml的诱变浓度对孢子致死率达79.92%,具有较好的诱变效果;初筛摇瓶产量比出发菌株提高20%以上的菌株有54株,占菌株的54%;连续3批进一步摇瓶复筛,并对复筛结果进行分析,发现116-5.8-64菌株所产南昌霉素活性单位高达9571.4mg/ml,比出发菌株提高了116.7%,差异达到极显著水平。
Different dosages of NTG were used to treat the spore suspension of nanchangmycin-producing Streptomyces nanchangensis 80-5.3- 116. The results showed that 500 μg/ml mutagenization dosage led to 79.92 % spore death rate had good mutagenic effect. Under this circumstancet, 54 spore mutants were obtained, yields of which were 20 % higher than that of the parent strains and covered 54 % in all in the pre-screening. Through continuous 3 batches fermentment, the yield of strain 116-5.8-64 was 9571.4 mg/ml, which was 116.7 % higher than that of the parent strains, and the difference between them was extremely significant.
