HPLC determination of mangiferin content in commercial Rhizoma Anemarrhenae

目的:建立知母药材中芒果苷的 HPLC 含量测定方法。方法:采用 Elite-ODS 色谱柱(4.6 mm×150 mm,5μm),以甲醇-0.1%磷酸溶液(27:73)为流动相,流速为1.0 mL·min^(-1),检测波长为316 nm,采用外标法定量测定。结果:芒果苷在0.214～1.07μg范围内线性关系良好(r=0.9997)。低、中、高3个浓度的平均回收率(n=3)分别为99.75%,98.47%,96.96%;RSD 分别为0.86%,0.91%,0.56%。结论:该方法准确可靠,可用于知母药材质量控制。

Objective： To establish an HPLC method for the determination of mangiferin content in Rhizoma Anemarrhenae. Method： An external standard method with Elite - ODS column （4.6 mm × 150 mm, 5 μm） and methanol -0. 1% phosphoric acid solution（27：73 ） as mobile phase was adopted. The wavelength for the detection was 316 nm and the flow rate was 1.0 mL · min^-1. Results：The linear range for the determination of mangiferin was 0. 214 - 1.07 μg（ r = 0. 9997 ）. The average recoveries（ n = 3 ） of low, middle, and high concentration were 99.75% ,98.47% ,96. 96% ,and RSDs were 0. 86% ,0. 91% ,0. 56% ,respectively. Conclusion：The method is accurate and can be used for the quality control of Rhizoma Anemarrhenae.
