食管鳞状细胞癌bFGF,TSP-1的表达与微血管密度的关系

The relationship between the expression of basic fibroblast growth factor,thrombospondin-1 and the microvessel density in esophageal squamous cell carcinoma

摘要 目的探讨食管鳞状细胞癌组织中碱性成纤维细胞生长因子(bFGF)、凝血酶敏感蛋白-1(TSP-1)和微血管密度(MVD)表达及其意义。方法采用免疫组化方法对46例病人食管鳞状细胞癌组织常规石蜡包埋切片分别检测 bFGF、TSP-1和 MVD。结果食管鳞状细胞癌组织 bFGF、TSP-1阳性率分别为60.9%(28/46)和37.0%(17/46),MVD 定量(56.59±28.97)/mm^2。T_3期肿瘤组织 bFGF 阳性表达率明显高于 T_2期者。bFGF 阳性表达的肿瘤组织 MVD 明显少于 bFGF 阴性表达的肿瘤组织[(49.51±24.73)/mm^2对(67.61±32.23)/mm^2,P<0.05)],T_3期、TNMⅢ期肿瘤组织 TSP-1阳性表达率明显高于 T_2期及 TNMⅡ期者。肿瘤细胞分化Ⅲ级者 MVD 明显高于Ⅰ、Ⅱ级者[(85.38±37-88)/mm^2对(41.35±20.32)/mm^2,P<0.01;(85.38±37.88)/mm^2对(54.28±23.43)/mm^2,P<0.01],伴有区域淋巴结转移组MVD 明显高于无淋巴结转移组[(65.38±32.33)/mm^2对(47.00±21.66)/mm^2,P<0.05]。结论 bFGF、TSP-1表达以及 MVD 计数与食管鳞状细胞癌组织的生物学行为有密切关系,bFGF 是调节食管鳞状细胞癌组织血管生成的重要因子。 Objective To study the expression of basic fibroblast growth factor (bFGF), thrombospondin-1 (TSP-1 ) and the microvessel density (MVD) and their relationship in esophageal squamous cell carcinoma. Methods Forty-six specimens of resected, paraffin embedded esophageal specious of squamous cell carcinoma were examined by immunohistochemical technique for the MVD and expression of bFGF and TSP-1. Results Of the 46 patients, the positive rate Of bFGF and TSP-1 were 60.9% (28/46) and 37.0% (17/46), respectively. The mean value of MVD was (56.59 ± 28.97)/mm^2. the depth of invasion had a significantly positive correlation with positive expression of bFGF and TSP-1. TNM stage had a significantly positive correlation with positive expression of TSP-1. the value of MVD was clesely associated with histological grade, lymph node metastasis and the expression of bFGF. Conclusion bFGF, TSP-1 and tumor microvessel play important roles in development and progression of esophageal squamous cell carcinoma. bFGF is a important regulator of tumor angiogenesis.
