银杏叶提取物对沙土鼠脑缺血再灌注损伤后SOD、CAT及MDA的影响 被引量:4

摘要 目的观察银杏叶提取物(EGb)对沙土鼠脑缺血再灌注损伤后氧自由基代谢的影响及该影响是否存在剂量依赖性。方法将健康成年沙土鼠随机分为四组,即假手术组、脑缺血再灌注组及EGb两个剂量组(50mg/kg、100mg/kg)。各组沙土鼠均缺血5min(假手术组除外),根据再灌注时间不同又分为3、6、12、24、72h亚组,每亚组各4只沙土鼠。分别于缺血前30min及缺血后即刻对GBE组给予银杏达莫注射液腹腔内注射,假手术组和缺血再灌注组给予等体积生理盐水。用比色法测量脑组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及丙二醛(MDA)的含量。结果大剂量EGb组与假手术组SOD、CAT、MDA水平相近,与缺血再灌注组比较差异显著,小剂量EGb组上述指标与缺血再灌注组水平相近。结论银杏叶提取物能有效减轻沙土鼠脑缺血再灌注损伤后氧自由基连锁反应所致的脑损伤,且存在明显的剂量依赖性。 Objective: To investigate the effect of extract of leave Gingko biloba on metabolism of oxygenic free radicals after cerebral ischemia reperfusion injury in Mongolian gerbil and to observe the influence whether or not shows a dose - depended manner. Methods: Adult Mongolian gerbil were divided into 4 groups randomly, including sham group, control group, 50mg/kg GBE group and 100mg/kg GBE group. Each group has five time points(3,6, 12,24,72h) according to the time of reperfusion after 5 min' s cerebral ischemia (except sham group). There are 4 Mongolian gerbils each time point. The rats received intraperitoneal injection of EGb(50mg/kg and 100mg/kg respectively)30 minutes before ischemia and injected again after ischemia immediately. The same volume of saline was injected intraperitoneally in group of sham and control. Measured SOD, CAT and MDA with colorimetric method. Results: There was no difference between 100mg/kg EGb group and sham, the same with 50mg/kg EGb group and control. Compared with control, 100mg/kg EGb group was better. Conclusion: EGb may alleviate cerebral injury caused by oxygenic free radicals with obvious dose - depended effect after ischemia reperfusion injury.
